Based on how this question was answered, we are to assume extroverts and shy people are not negations of each other? If so, should we assume most premises will include explicit words such as "not" to indicate the negation?

Yes, assume that shy people and extroverts aren't each others'
negations. There aren't any hard and fast rules, but in general,
premises will include a word like "not" to indicate the negation.
